شناسایی و آنالیز توالی cDNA کد کننده پروتئین شوک حرارتی 90 کیلو دالتون (Hsp90) در ماهی سفید (Rutilus frisii kutum)
گروهی مهمی از چاپرونها که به پروتئینهای شوک حرارتی (Hsp) معروف هستند، وظیفه اصلی آنها نظارت بر تاخوردگی پروتئینهای درون سلولی است. پروتئین Hsp90 که فراوانترین پروتئین در سلولهای یوکاریوتی نیز است )حدود ٪2-1 از کل پروتئینهای سلول) علاوه بر نقشی که ذکر شد در امور دیگر مانند ایجاد کمپلکس پروتئینهای مسیرهای انتقال پیام، حائز اهمیت است. در این پژوهش Hsp90 گونه Rutilus frisii kutum ماهی سفید که گونه بومی ایران است مورد بررسی قرار گرفت. از آنجایی که اطلاعاتی در مورد توالی ژن hsp90 در ماهی سفید در دست نبود ابتدا توالی این ژن به کمک تکنیک PCR و توالییابی محصول PCR به دست آمد. نتایج نشان داد که این توالی همولوژی بالای 97 درصد با hsp90 گونههای Cyprinus carpio و Gobiocypris rarus داشت. طول کامل ژن hsp90 مورد مطالعه 2181 نوکلئوتید مطابق با 727 ریشه آمینو اسید به دست آمد.

غیرفعالسازی ویروس Rhabdovirus carpio (ویروس ویرمی بهاره کپور) با استفاده از نانوذرات نقره در سلولهای رده EPC
در میان عوامل بیماریزای آبزیان، نقش ویروسها از همه بارزتر بوده، خسارات وارده به علت درمانناپذیری، سرایتپذیری شدید و تشخیص دشوار مهمتر از سایر عوامل بیماریزا است. در این مطالعه اثر ضدویروسی نانوذرات نقره بر ویروس Rhabdovirus carpio، عامل ویرمی بهاره کپور (SVCV)، از طریق مواجهسازی سلولهایEPC آلوده به ویروس با نانوذرات نقره بررسی شد. در ابتدا سمیت سلولی نانوذرات نقره بر روی تیره سلولی تعیین شد و از غلظتهای پایینتر از غلظت نیمه سمی (CC505/0) برای مواجهسازی استفاده شد. مواجهسازی سلولهای آلوده به ویروس با نانوذرات نقره به دو روش تلقیح همزمان و تلقیح با تاخیر چهار ساعته انجام شد. سلولهای تلقیح شده، به مدت شش روز، از نظر بروز آثار آسیب سلولی و تغییرات جمعیت سلولهای زنده بررسی شدند. بر اساس نتایج، غلظت نیمه سمی نانوذرات نقره در سلولهای EPC mg/L62 تعیین شد. همچنین، میزان تلفات سلولی در دو تیمار تلقیح همزمان و تاخیری به ترتیب %39 و %26 بود، در حالی که در این مدت %100 سلولهای شاهد آلوده با ویروس خالص تخریب شدند. بر اساس این مطالعه، نانوذرات نقره در غلظتهای با سمیت کم از قابلیت مناسبی در غیرفعالسازی ویروس ویرمی بهاره کپور برخوردار است و میتواند به عنوان یک عامل ضدویروسی در مهار عفونت SVCV در آبزیپروری مدنظر قرار گیرد.

بررسی اثر تجویز خوراکی سطوح مختلف پروبیوتیک Lactobacillus casei بر برخی آنزیمهای گوارشی ماهی قزلآلای رنگینکمان انگشتقد
پروبیوتیکها مکملهای غذایی زندهای هستند که از طریق بهبود بار میکروبی روده میزبان، تاثیرات سودمندی را در آن ایجاد کرده، سبب بهبود جذب مواد غذایی از روده میشوند. هدف از این مطالعه بررسی اثر تجویز خوراکی سطوح پروبیوتیک Lactobacillus casei (PTCC1608) بر برخی آنزیمهای گوارشی ماهی قزلآلای رنگینکمان است. از این رو، 4800 قطعه بچه ماهی قزلآلا، با وزن متوسط 5/5±6/32 گرم در چهار تیمار با سه تکرار به صورت تصادفی تقسیم شدند (هر تکرار با 400 قطعه ماهی). ماهیان تیمارهای A، B و C به ترتیب با جیرههای غذایی حاوی 106×5، 107×5 و CFU/mL108×5 L. casei به مدت 60 روز غذادهی شدند. یک تیمار نیز به عنوان تیمار شاهد با شرایط کاملا مشابه اما بدون اضافه کردن باکتری به خوراک در نظر گرفته شد. با اندازهگیری فعالیت آنزیمهای آلفا آمیلاز، ترپسین و کموتریپسین در روز 60 مشخص شد تیمارهایی که از پروبیوتیک با جیره حاوی CFU/mL106×5 L. casei تغذیه شده بودند در مقایسه با گروه شاهد اختلاف معنیدار داشتند (05/0P˂). اما در سایر تیمارهایی که با غلظتهای دیگر پروبیوتیک تغذیه شده بودند، اختلاف معنیداری در فعالیت این آنزیمها نسبت به گروه شاهد مشاهده نشد (05/0P>). همچنین فعالیت آنزیمهای آلکالین فسفاتاز، لیپاز و پروتئاز در تیمارهای که از پروبیوتیک با غلظتهای 106×5 و CFU/mL107×5 استفاده شده بود، افزایش معنیداری را نسبت به شاهد نشان داد (05/0P˂). بر اساس نتایج این مطالعه، افزودن پروبیوتیک L. casei با غلظت CFU/mL106×5 به خوراک ماهی قزلآلای رنگینکمان انگشتقد، به عنوان مناسبترین غلظت، باعث بهبود فعالیت آنزیمهای گوارشی ماهی قزلآلا میشود، ولی غلظتهای بالاتر آن، اثرات منفی بر ماهی قزلآلا دارد.

برآورد نسبی پتانسیلهای ژنتیکی خنثی و تنوع سازشی در جمعیتهای پرورشی کپور نقرهای (Hypophthalmichthys molitrix) کشور
در دهه اخیر، مطالعه نشانگرهای خنثی در برابر انواعی که نماینده تنوع سازشی هستند مورد اقبال قرار گرفته است که شاخصترین آنها، مجموعه ژنهای MHC است. در مطالعه حاضر نشانگر MHC به همراه 8 نشانگر ریزماهواره بر روی 138 قطعه کپور نقرهای در استانهای مختلف کشور شامل گلستان، مازندران، گیلان، خوزستان و نمونههای وارداتی از کشور چین، مورد بررسی قرار گرفت. میانگین غنای اللی، هتروزیگوسیتی مورد انتظار و شاخص شانون به ترتیب برای ریزماهوارهها 04/5، 682/0 و 32/ 1 و جایگاه MHC 21/4، 674/0 و 23/1 به دست آمد. آنالیز AMOVA در جایگاههای ریزماهواره و MHC به ترتیب 2 و 3 درصد از واریانس تنوع ژنتیکی را به تفاوتهای بینجمعیتی اختصاصداد. با اجرای آزمون وضعیت تعادل در جمعیتهای مورد مطالعه با استفاده از مدل SMM، بروز تنگنای ژنتیکی تایید نشد. با توجه به افزایش معنیدار هتروزیگوسیتی در هر دو نوع جایگاه در مقایسه با مدل هاردی- وینبرگ و برتری نسبی MHC در ایجاد تمایز بین جمعیتها میتوان نقش بهگزینی تعادلی را به این جایگاه نسبتداد. جمعبندی نهایی مبین آن است که تنوع اللی جایگاههای مذکور در جمعیتهای مورد بررسی طی چندین دوره تکثیر و پرورش حفظ شده است. غنای اللی بالاتر جایگاه MHC-DAB در میان نمونههای چینی در کنار سطح هتروزیگوسیتی و تنوع بالای درونجمعیتی میتواند بهعنوان نقطهقوت این گروه بهشمارآید.

اثرات جایگزینی روغنهای گیاهی با روغن ماهی در جیره غذایی بر آنزیمهای کبدی سرم و بافت کبد ماهی قزلآلای رنگینکمان (Oncorhynchus mykiss)
این مطالعه برای ارزیابی تاثیر اسیدهای چرب بر بافت کبد و همچنین تغییرات آنزیمهای کبدی سرم خون ماهی قزلآلای رنگینکمان در هنگام جایگزینی کامل روغن ماهی با روغنهای گیاهی انجام شد. برای این منظور چهار تیمار شامل تیمار 1 (شاهد): جیره حاوی روغن ماهی، تیمار 2: جیره حاوی روغن سویا، تیمار 3: جیره حاوی روغن آفتابگردان و تیمار 4: جیره حاوی روغن کانولا، در نظر گرفته شد. 25 قطعه ماهی با وزن متوسط 13/1±12/65 گرم در هر حوضچه (برای هر تیمار 2 حوضچه) به صورت تصادفی توزیع و به مدت 50 روز تغذیه شدند. با توجه به نتایج به دست آمده، اختلاف معنیداری از لحاظ وزن نهایی بین تیمارهای مختلف مشاهده نشد. مقادیر آنزیمهای کبدی (AST، ALT و LDH) در تیمار حاوی %100 روغن ماهی به طور معنیداری بیشتر از سایر تیمارها بود. بافت کبد در تمامی تیمارها ضایعاتی از قبیل دژنرسانس چربی، هیپرتروفی، نکروز سلولی، پرخونی و خونریزی داشت. این ضایعات در تیمار شاهد بیشتر بود. با توجه به نتایج به دست آمده، اکسیداسیون روغن ماهی در جیره باعث آسیب دیدن کبد و بالا رفتن مقادیر آنزیمهای کبدی سرم خون میشود. در نتیجهگیری نهایی میتوان بیان کرد که حداقل بخشی از روغن جیره میتواند از منابع گیاهی تامین شود. همچنین در هنگام استفاده از روغن ماهی در جیره باید به کیفیت آن توجه زیادی شود.

کارآیی مواد زیستفعال تولیدی از باکتری Lactococcus lactis همراه با اسانسهای گیاهی، نمک و اسید استیک جهت کنترل Listeria monocytogenes در مدل مایع و گوشت چرخ شده Hypophthalmichthys molitrix
استفاده از تکنولوژی نگهداری زیستی به کاهش استفاده از نگهدارندههای شیمیایی برای تولید غذاهای طبیعی با حفظ خواص ارگانولپتیکی کمک میکند. در این پژوهش، تغییرات میکروبی و کیفیت حسی گوشت چرخ شده ماهی فیتوفاگ تحت تاثیر اسانسهای مرزنجوش، پونه کوهی و شوید، نایسین و اسید استیک طی نگهداری در یخچال به مدت 12 روز مورد بررسی قرار گرفتند. تیماربندیها شامل تیمار شاهد، گوشت تلقیح شده با اسانسهای پونه کوهی، مرزنجوش و شوید در غلظتهای 2/0، 4/0، 6/0، 8/0 و 1 درصد، نمک، نایسین و تیمارهای ترکیبی (مرزنجوش- پونه کوهی، پونه کوهی- اسید استیک و مرزنجوش- اسید استیک) بودند. با توجه به نتایج، مرزنجوش بیشترین قطر هاله عدم رشد و تاثیر را روی Listeria monocytogenes از خود نشان داد. همچنین میزان MIC این اسانس μL/mL5/2 و MBC در حد μL/mL75/3 گزارش شد. شاخص حسی کلیه نمونهها طی نگهداری، کاهش پیدا کردند که در نمونه شاهد بیشتر از نمونههای تیمار شده با اسانس بود. در آزمایش کدورتسنجی، اثر تیمار و زمان معنیدار بود و همه تیمارها نسبت به شاهد تفاوت معنیداری نشان دادند. کمترین کدورت مربوط به اسید استیک و بیشترین کدورت مربوط به تیمار شاهد بود. بنابراین، کاربرد توام نایسین با اسانسهای گیاهی برای تامین ایمنی غذایی، مطلوب و مناسب است.

بررسی اثر محافظتی ویتامین C بر شکستگی DNA در سلولهای آبشش و کبد ماهی قزلآلای رنگینکمان (Oncorhynchus mykiss) در مواجهه با غلظتهای مختلف نانوذره اکسید روی (ZnO)
این مطالعه به بررسی اثر آلاینده نوظهور نانوذره اکسید روی (ZnO) بر روی بافت کبد و آبشش ماهی قزلآلای رنگینکمان (Oncorhynchus mykiss) و به دنبال آن بررسی اثر ایمنی ویتامین C بر شکستگی DNA پس از اعمال نانوذره پرداخته است. برای این منظور از ترکیب سطوح مختلف نانوذره روی (0، 40 و 80 میلیگرم در لیتر) و ویتامینC (0، 400 و 800 میلیگرم در کیلوگرم) استفاده شد (9 تیمار هر یک با سه تکرار) و ماهیان با میانگین وزنی 10±170 گرم به مدت 10 روز تحت تیمار با جیرههای آزمایشی قرار گرفتند. نمونهگیری در روزهای پنجم و دهم آزمایش از بافت کبد و آبشش صورت گرفت. نمونههای مورد نظر پس از جداسازی در میکروتیوپهای حاوی الکل %96 نگهداری شدند. استخراج DNA به روش شکستگی DNA با الکتروفورز انجام گرفت، سپس با ژل الکتروفورز اجرا شد و در پایان با دستگاه ژلداک عکسبرداری شد. درصد شکستگیDNA با استفاده از میانگین وزنی تعیین شد. بر طبق نتایج، اثر غلظت ZnO و ویتامین C و اثر متقابل زمان- غلظت بر روی سطح آسیب DNA در بافت کبد و آبشش بین تیمارهای مختلف کاملا معنیدار بود. بیشترین آسیب در بافت کبد و آبشش در غلظت80 میلیگرم بر لیتر نانوذره اکسید روی در روز پنجم مشاهده شد. همچنین در هر دو بافت در روز دهم میزان شکستگی DNA کاهش یافت. به علاوه، حضور ویتامین C موجب کاهش شکستگی DNA شد.
