اثرات دیازپام و نورتریپتیلین بر تغییرات رفتاری و افسردگی ناشی از استرس مزمن در ماهی گورخری (Danio rerio)

چکیده

افسردگی یک بیماری ذهنی است و طبق آمار‌ها هر روز جدی‌تر می‌شود که نشان از نیاز روز‌افزون به مطالعه بر روی مدل‌های افسردگی و تولید دارو‌های ضد‌افسردگی جدید دارد. هدف این پژوهش، ارزیابی تاثیرات منفرد و ترکیبی دارو‌های دیازپام و نورتریپتیلین بر شاخص‌‌های رفتاری و بیوشیمیایی در ماهی گورخری (Danio rerio) بوده است. در این مطالعه، ۳۸۴ ماهی گورخری ابتدا به دو دسته کلی شاهد (بدون عوامل استرس‌زا) و UCS (با استرس‌زا) تقسیم شدند و درون هر دسته نیز ماهی‌ها به تیمار‌های شاهد، دیازپام، نورتریپتیلین و دیازپام + نورتریپتیلین تقسیم شدند (هشت تیمار با سه تکرار). تعداد ماهی‌های هر تیمار، ۱۶ قطعه و هر تیمار نیز دارای ۳ تکرار بود. برای ایجاد افسردگی در ماهی‌ها از پروتکل Unpredictable Chronic Stress (UCS) استفاده شد. آزمایش‌های رفتاری با آزمایش مخزن جدید (NTT) و آزمایش‌های بیوشیمیایی با اندازه‌گیری کورتیزول تام بدن انجام شد. دیازپام و نورتریپتیلین موجب بهبود قابل توجه و معنی­داری در شاخص‌های رفتاری شامل افزایش معنی‌دار در مدت زمان حضور در سطح بالایی و تعداد انتقال به سطح بالایی و کاهش معنی‌دار مسافت کل طی شده و پیشگیری از اثرات بیوشیمیایی افسردگی با کاهش معنی­دار سطوح کورتیزول (از ۰۷/۰±۰۱/۱ به ۱۶/۰±۳۸/۰ نانوگرم) شدند. در این مطالعه داده‌های پژوهش‌های مشابه پیشین در این زمینه تایید شد که نشان می‌دهد پروتکل UCS موجب القای تغییرات رفتاری و نوروآندوکرین در ماهی گورخری می‌شود که با مصرف دیازپام و نورتریپتیلین، می‌توان از آنها پیشگیری کرد. همچنین این مطالعه استفاده از ماهی گورخری را به عنوان مدل در مطالعات تاثیرات رفتاری و فیزیولوژیکی استرس تقویت کرد
