تعیین کیسپپتین ماهی سفید دریای خزر و تاثیر آن بر هورمون‌های تولیدمثلی

چکیده

ماهی سفید دریای خزر (Rutilus kutum) از نظر تجاری و غذایی مهم‌ترین ماهی استخوانی حوضه جنوبی دریای خزر است که برای جلوگیری از انقراض این گونه با ارزش از تولیدمثل القایی برای هم‌زمان‌سازی و افزایش میزان هماوری در شرایط پرورشی استفاده می‌شود. مطالعات پیشین نشان داد که کیسپپتین که یک پروتئین فوق محرک در محور غدد ترشح ‌کننده هورمون‌های جنسی است، منجر به بلوغ جنسی و گامتوژنز می‌شود، اما تاکنون توالی کیسپپتین 1 و 2 ماهی سفید مشخص نشده است و متناظرا  قابلیت این پپتید از پروتئین (Kiss1) در تولیدمثل مورد بررسی قرار نگرفته است. در مطالعه حاضر توالی کیسپپتین 10 در ماهی سفید به روش CLUSTALW و با طراحی آغازگر از توالی Kiss1 ماهی‌های هم‌خانواده ماهی سفید تعیین شد. برای دست‌یابی به بهترین غلظت پپتید Kiss1 از غلظت‌های 5، 25، 50 و 100 میکروگرم در کیلوگرم استفاده شد و غلظت 17-α هیدروکسی پروژسترون و 17-β استرادیول پلاسما مورد بررسی قرار گرفت. نتایج این مطالعه نشان داد که کیسپپتین 10 سنتزی با غلظت بهینه 100 میکروگرم در کیلوگرم می‌تواند به عنوان گزینه مناسب برای القا تخم‌ریزی در ماهی سفید معرفی شود.
